Larpose Tablet contains an active ingredient called lorazepam which belongs to a class of medications known as benzodiazepines. This medicine acts on the brain and nerves (central nervous system). It

is an intermediate-acting tranquilliser that interferes with the natural state of certain neurotransmitters called gamma-aminobutyric (GABA). Lorazepam binds to these GABA receptors and enhances the GABA enhanced synaptic inhibition, responsible for its calmness and sedation effects.

No, avoid taking Larpose Tablet during pregnancy since it is a category D medicine. There are certain teratogenic or foetus-harming substances in the medicine. Therefore the use of this medicine during pregnancy is not recommended. If a benzodiazepine is required, a medicine other than Larpose can be preferred. Consult a doctor or a gynaecologist before starting any benzodiazepine during pregnancy. Never self prescribe, or self medicate.

It can also cause panic attacks, dizziness or feeling faint, memory loss, hallucinations, feeling stiff and unable to move easily, feeling very warm, convulsions (sudden uncontrolled shaking or jerking of the body) and oversensitivity to light, sound and touch.

The effect of Larpose 2mg Tablet on weight gain or loss is not known.

If you have a history of depression then it may increase the risk of developing depression again. Larpose 2mg Tablet should not be used alone in depressed patients because it may cause suicidal tendencies in such patients.

Though it is rare, but use of Larpose 2mg Tablet can cause memory impairment, which may be more apparent in old age patients.

What is this drug used for?

• It is used as a substitute for thyroid hormone.
• Used to treat thyroid cancer.
• Used to diagnose thyroid problems.

What should I tell my doctor BEFORE taking this drug?

• If you have an allergy to this drug, any of its ingredients, other drugs, foods or substances. Tell your doctor about your allergies and how they have manifested.

• If you have high blood sugar (diabetes), this drug can sometimes raise your blood sugar even more. Discuss with your doctor how you can keep your blood sugar under control.
• Check your blood sugar as directed by your doctor.
• Perform blood tests as directed by your doctor. Consult with your doctor.
• Taking this drug at excessively high doses can cause broken bones (osteoporosis). The risk may be higher after menopause. If you want to know if you are at increased risk for broken bones, or if you have any questions, check with your doctor.
• If you are 65 years of age or older, use this drug with caution. You may experience more side effects.
• In some cases, the drug may affect the growth rate in children and adolescents. They may need regular growth rate checks. Consult your doctor.
• Tell your doctor if you are pregnant, planning to become pregnant, or breastfeeding. The benefits and risks for you and your child will need to be discussed.

What side effects should I report to my doctor immediately?

WARNING. In rare cases, this drug can cause serious and sometimes deadly side effects in some patients. Contact your doctor or seek medical attention right away if you have any of the following signs or symptoms that may be associated with serious side effects:

• Signs of an allergic reaction such as rash, hives, itching, red and swollen skin with blisters or peeling, possibly in combination with fever, wheezing or wheezing, tightness in the chest or throat, difficulty breathing, swallowing or speaking, unusual hoarseness, swelling in the mouth, face, lips, tongue or throat.

What to do if you suspect that you are having side effects from a vaccine? – vaktsineeri.ee

The vast majority of vaccinees do not experience any particular side effects after being vaccinated against COVID-19. But if you suspect you have some more serious side effects, you should definitely contact your family doctor.

Reports of side effects in Estonia are collected by the State Medicines Board. The collected information on side effects received by the Medicines Board is published weekly on the website of the agency www.ravimiamet.ee

The most common post-vaccination reaction lasting a couple of days after vaccination is, for example, soreness or tenderness at the injection site, flu-like symptoms such as dizziness, headache, slight fever, drowsiness, etc.

These reactions can be relieved with pain medication or a cold compress if needed. In most cases, these symptoms resolve within a couple of days. However, if the discomfort persists for longer, or if you feel really bad after the vaccination, contact your family doctor. All post-vaccination complaints must be reported to the family doctor. For additional description of the safety profile of COVID-19 vaccinesall health care workers should report all systemic and serious (including local) side effects that may be associated with the vaccine. If the doctor reports this to the Department of Medicines, then the person who received the vaccine does not need to do this separately.

You can also report on the website of the Medicines Board those reactions that, according to the doctor, are not related to the vaccine. The Department collects information in Estonia related to the use of vaccines (and other medicines) and the occurrence of side effects in order to identify new side effects or to clarify changes in the severity and frequency of already known side effects. And also whether a new message can better minimize the risks.

Submitted information requires follow-up

The Department of Medicines analyzes the data received in the report and makes an initial assessment of whether these health conditions are related to a particular drug or vaccine, or if the problems were caused by other causes. If reports are received containing information about serious reactions, the Department of Medicines always asks the patient for the contact details of the doctor in order to obtain medical confirmation and additional data, for example, whether the patient has comorbidities or other risk factors, what tests and studies were performed.

The data collected in Estonia are transferred to the side effects database of the European Medicines Agency. The safety assessment of vaccines is carried out by the Risk Assessment Committee of the European Medicines Agency and the Committee for Medicinal Products for Human Use, which also includes experts from the Estonian Medicines Board. Experts regularly evaluate all new possible side effects by analyzing large amounts of data, taking into account the frequency of manifestation of the disease / health disorder in the general population. Similarities found in the general population and more frequent than previously thought reactions identified during the analysis lead to the assertion of an association between the vaccine and the reaction to it.

The Estonian and European side effects database (adrreports.eu) cannot be used to decide on the safety of any vaccine or drug, because it contains all reports that are received about drugs and vaccines, and only after that they are evaluated and control.

The Centers for Disease Control and Prevention (CDC) Vaccine Adverse Event Reporting System (VAERS) database is compiled in the same way. Reports enter the database without control and there are reports of cases where there is no connection between the vaccine, the drug and the reaction.

A lot of people are being vaccinated against the coronavirus infection in a very short time. This means a noticeably greater than usual probability that the manifestation of a particular disease or its aggravation occurs at the time of vaccination, but is not a consequence of it.

Knowledge is growing

When assessing safety, it is important to take into account the frequency of manifestation of certain diseases in the general population. If, when evaluating data from all over Europe, it turns out that the vaccinated as a whole get a certain disease more often, or, for example, it manifests itself in a certain age group more often than in the general population, then this is a very significant danger signal. If the danger signal is confirmed, it is subjected to a separate assessment, and if a clear justification for the relationship is determined, then this is included in the instructions for use of the drug as a side effect.

Thus, myocarditis was added as a possible side effect to mNRK vaccines (Pfizer/Corminatry and Moderna/Spikevax), and thrombosis syndrome with thrombocytopenia was added to adenoviral vaccines (Janssen, AstraZeneca/Vaxzevria) as a possible side effect.
